Senior Scientist – CRISPR Specificity & Genomic Safety
Profluent · Emeryville
Job description
About the role
Profluent is seeking a senior scientist to design and execute experimental workflows that evaluate the specificity, safety, and developability of its AI‑designed CRISPR base‑editing platform. The role will focus on establishing robust off‑target discovery methods and scaling them across multiple gene‑editing modalities.
Key responsibilities
- Define and lead the strategy for base‑editor specificity assessment, including CHANGE‑Seq‑BE and related biochemical, cell‑based, and targeted sequencing approaches.
- Develop, optimize, benchmark, and apply orthogonal methods to evaluate editing specificity for base editors, nucleases, and broader genomic integrity.
- Design and execute studies to identify, prioritize, and validate off‑target editing events, handling assay development, NGS library preparation, sequencing QC, and data interpretation.
- Generate high‑quality datasets that guide editor engineering, candidate selection, delivery conditions, and program development.
- Collaborate cross‑functionally with protein engineering, cell biology, bioinformatics, automation, and program teams.
Required profile
- PhD or equivalent experience in molecular biology, genetics, or related field.
- Demonstrated experience with CRISPR base‑editing platforms and off‑target assessment methods.
- Proven ability to develop and benchmark biochemical and cell‑based assays.
- Strong track record of interpreting NGS‑based specificity data.
- Experience working in interdisciplinary, fast‑moving research environments.
Required skills
- CRISPR base editing
- CHANGE‑Seq‑BE
- Next‑generation sequencing (NGS) data analysis
- Assay development and optimization
- Biochemical and cell‑based off‑target discovery
- Bioinformatics integration
- Automation of experimental workflows
What we offer
- Opportunity to shape AI‑driven genome editing evaluation.
- Collaboration with leading experts in machine learning, protein engineering, and genomics.
- Access to state‑of‑the‑art laboratory and computational resources.
